Understairs headroom is the usual constraint
The WC needs to sit where there is standing headroom, which is normally at the tall end of the understairs void. That determines the whole layout, and the low end is then best used for bespoke storage.
Ventilation is a requirement, not a preference
A cloakroom with no openable window needs mechanical extraction with an overrun. It is a building regulations matter and, in a small internal room, a very obvious one if it is missing.
Do not lose the whole understairs space
A well-planned cloakroom uses about two-thirds of the void. The rest can become drawers, a shoe cupboard or a cleaning store — worth doing while the space is open.
